Exosomes are nano size of complex biological membrane structure that are secreted by most cell types and function as mediators of intercellular communication and tissue regeneration. In this study, we hypothesized that exosomes released from osteogenic differentiation (D-EXO) of human adipose-derived stem cells (hASCs) may contain potential factors for promoting bone regeneration. Moreover, we fabricated D-EXO-loaded extracellular matrix (ECM) scaffolds for bone tissue engineering. In an in vitro study, the D-EXO showed low cytotoxicity and hASCs treated with D-EXO were more differentiated in comparison with control groups. In an in vivo study, the osteogenesis was partially induced in D-EXO-loaded ECM scaffold and ECM scaffold only groups. Our results indicate that D-EXO may regulate osteogenesis and D-EXO-loaded ECM scaffold may provide a cell-free therapy in bone tissue regeneration.
